1. A method of detecting a methylated cytosine of the invention comprises the steps of: hybridizing an oligonucleotide capable of hybridizing to a region containing a cytosine which is expected to be methylated in a sample DNA and having an abasic site at a position complementary to the cytosine to the sample DNA; oxidizing a methylated cytosine by reacting an oxidizing agent with the hybridized sample DNA formed in the previous step when a methylated cytosine is present therein; and detecting the oxidized methylated cytosine.

L'invention porte sur un procédé de détection d'une cytosine méthylée de l'invention comprenant les étapes consistant à : hybrider un oligonucléotide capable de s'hybrider à une région contenant une cytosine censée être méthylée dans un ADN échantillon et ayant un site abasique à une position complémentaire de la cytosine par rapport à l'ADN d'échantillon; oxyder une cytosine méthylée par réaction d'un agent oxydant avec l'ADN échantillon hybridé formé dans l'étape précédente lorsqu'une cytosine méthylée est présente dans celui-ci; et détecter la cytosine méthylée oxydée.
